ABSTRACT

ObjectiveTo explore the effect of Qishengwan on ileal flora during its treatment of Alzheimer's disease (AD) under the guidance of the theory of "interior-exterior relationship between heart and small intestine". MethodThe AD model was established by bilateral intraventricular injection of β-amyloid 1-42 (Aβ1-42). The rats were then randomly divided into the blank group, sham-operated group, model group, low-, medium-, and high-dose (5.6, 11.2,22.4 g·kg-1·d-1) Qishengwan groups, and donepezil (0.46 mg·kg-1·d-1) group. After medication for 28 successive days, the spatial memory ability of rats was observed in water maze test, and the levels of Aβ1-42, nuclear transcription factor-κB (NF-κB), tumor necrosis factor-α (TNF-α), and interleukin-6 (IL-6) in the hippocampus were analyzed by enzyme-linked immunosorbent assay (ELISA). Additionally, the contents of the ileum were collected and subjected to 16SrRNA-sequencing analysis for figuring out the changes in ileal flora. ResultCompared with the blank group and sham-operated group, the model group exhibited significantly reduced stay time in the target quadrant and number of target quadrant and platform crossings (P<0.05, P<0.01) and elevated Aβ1-42 content in the hippocampus (P<0.01) and central inflammatory factors NF-κB, TNF-α, and IL-6 (P<0.05, P<0.01). Compared with the model group, Qishengwan at each dose significantly alleviated the impaired spatial memory function (P<0.05, P<0.01), improved the deposition of Aβ1-42 in the hippocampus of rats (P<0.05, P<0.01), and reduced the expression of central nervous system inflammatory factors (P<0.05, P<0.01), thus exerting a good therapeutic effect on AD rats. The 16SrRNA-sequencing analysis results showed that the structure of the ileal flora in the model group was significantly separated from those in the blank group and sham-operated group. The abundance of Lachnospiraceae NK4A136 group was significantly increased (P<0.01), while that of Escherichia-Shigella was reduced (P<0.05, P<0.01). Qishengwan at each dose significantly changed the ileal flora structure and regulated the relative abundance of Lachnospiraceae NK4A136 group, Escherichia-Shigella, and Ruminococcaceae. ConclusionQishengwan has a positive therapeutic effect on AD. It can significantly enhance the memory and cognitive abilities in AD rats, which may be related to its regulation of the structure of rat ileal flora and the relative abundance of Lachnospiraceae NK4A136 group, Escherichia-Shigella, and Ruminococcaceae, the attenuation of the central neuroinflammatory response, and the reduction of central Aβ1-42 deposition.

ABSTRACT

Objective To observe the clinical efficacy of acupuncture plus hyperbaric oxygen in treating depression after cerebral stroke.Method A total of 120 patients with depression after cerebral stroke were randomized into a treatment group of 60 cases and a control group of 60 cases. The treatment group was intervened by acupuncture plus hyperbaric oxygen, while the control group was intervened by Fluoxetine. Before the treatment, and respectively after 14-d and 28-d treatments, the patients were evaluated by Hamilton Depression Scale (HAMD) and National Institute of Health Stroke Scale (NIHSS), and the therapeutic efficacies were compared between the two groups. Result The treatment group was significantly superior to the control group in comparing therapeutic efficacy (P<0.01). After 1 course of treatment, the HAMD and NIHSS scores dropped significantly in the treatment group (P<0.01); the scores in the control group showed a declining tendency but without statistical significances (P>0.05). After 2 treatment courses, the HAMD and NIHSS scores were significantly different from those before the treatment in both groups (P<0.01), and the differences were more significant in the treatment group (P<0.01).Conclusion Acupuncture plus hyperbaric oxygen is effective in treating depression after cerebral stroke, and it can obviously improve the neural function.

ABSTRACT

OBJECTIVE: To establish an HPLC-UV-ESI-MSn method for the study of impurity profile of amoxicillin and clavulanate potassium tablets. METHODS: Agilent 1100 LC/MSD Trap liquid chromatography-mass spectrometry was used, and the column was Shim pack CLC-ODS RP18(4.6 mm×250 mm, 5 μm). The mobile phase A was 20 mmol·L-1 ammonium acetate (pH adjusted to 6.0), and the mobile phase B was 20 mmol·L-1 ammonium acetate-acetonitrile (20∶80) (pH adjusted to 6.0). Gradient elution was performed at a flow rate of 1.0 mL·min-1. ESI source was used. Positive and negative ion scan was conducted with a scanning range of m/z 100-1 500. The nebulizing pressure was 275.8 kPa, dry gas flow was 9 L·min-1, and post-column diversion ratio was 1∶5. Some related substances were identified by comparing the retention time in the chromatography, [M+H]+ spectrum and MS2 spectrum with those of the reference substances, while the others which do not have reference substances were deduced or speculated by analyzing the MS2 or MSn fragmentation with the help of a rule summarized from the MS2 fragmentation of amoxicillin, clavulanic acid and system suitability impurity reference substances. RESULTS: A total of 15 related substances were separated and characterized including nine known impurities like amoxicilloic acid, amoxicillin dimer, etc. and six unknown impurities. CONCLUSION: The method can be applied in the identification and qualitative analysis of the related substances in amoxicillin and clavulanate potassium tablets and is helpful for the quality control and optimization of the synthetic process.

ABSTRACT

BACKGROUND:Spleen deficiency has varying impact on the immune system and digestive system of the human body, and can also damage the normal function of the central nervous system as there is a close relationship between the spleen and brain. OBJECTIVE:To explore the effect of electroacupuncture on the proliferation and differentiation of neural stem cels in the rat hippocampus of spleen deficiency syndrome. METHODS:Sixty Sprague-Dawley rats were randomly divided into three groups: normal, control and electroacupuncture groups. The animal model of spleen deficiency syndrome was prepared in the control group and electroacupuncture group. Two weeks after modeling, rats in the electroacupuncture group were given electroacupuncture treatment, and the changes of body mass in normal rats and model rats were measured. Five rats from each group were taken to observe the histological changes of neural stem cels in the hippocampus at 1, 2, 3, 4 weeks after treatment. RESULTS AND CONCLUSION: The body mass of rats in the model group was significantly lower than that in the normal group at 2 weeks after modeling (P < 0.05). In the electroacupuncture group, the number of BrdU positive cels was significantly higher than that in the normal group and control group at 2, 3, 4 weeks after modeling (P < 0.05); the number of BrdU/Nestin positive cels was significantly higher than that in the normal group at 2, 3, 4 weeks (P < 0.05); the number of BrdU/glial fibrilary acidic protein positive cels was significantly higher than that in the normal group at 2 weeks (P < 0.05); and the number of BrdU/neuronspecific enolase positive cels was significantly higher than the normal group at 3 weeks (P < 0.05). In the control group, the number of BrdU/Nestin positive cels was significantly lower than that in the normal group and electroacupuncture group at 1 week after modeling, significantly lower than that in the electroacupuncture group at 2 and 4 weeks, and significantly lower than the normal group at 3 weeks (P < 0.05); the number of BrdU/glial fibrilary acidic protein positive cels was significantly lower than that in the normal group and electroacupuncture group at 1, 2, 4 weeks; and the number of BrdU/neuronspecific enolase positive cels was significantly lower than that in the normal group and electroacupuncture group at 1, 2 weeks. Taken together, these experimental findings show that electroacupuncture treatment effectively promotes the proliferation of hippocampal neural stem cels that are induced to differentiate into astrocytes and neurons.

ABSTRACT

Objective To investigate the correlation between the positive rate of high risk human papillomavirus (HPV) mRNA E6/E7 and cervical cancer,and provide evidence for the prevention and treatment of cervical cancer.Methods A total of 100 cervical cancer cases and 100 healthy controls were selected in our hospital from January 2015 to December 2015.The fluorescence quantitative PCR and pathological examination on HPV E6/E7 mRNA were carried out.The correlation between HPV E6/E7 mRNA and cervical squamous epithelial lesions were analyzed.Results In case group,the positive rate ofHPV E6/E7 mRNA was 76.0% (76/100).In control group,the positive rate was 13.0% (13/100).The positive rate in case group was significantly higher than that in control group,and the difference was statistically significant (x2=24.522,P<0.001).The positive predictive value and negative predictive value of the two groups were compared,and the difference was not significant (P>0.05).The positive rate of HPV E6/E7 mRNA was significantly higher than high-grade squamous intraepithelial lesion (SIL) rate (26.1%),low-grade SIL rate (17.6%) and atypical squamous cell hyperplasia rate (6.7%),the difference was statistically significant (x2=7.615,P=0.001;x2=9.114,P=0.001;x2=18.241,P<0.001).Conclusions The detection rate ofHPV E6/E7 mRNA in cervical cancer patients was high.And with the increased severity of cervical squamous epithelial lesions,the positive rate of HPV E6/E7 mRNA increased.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the regulatory effects of electroacupuncture with different acupoints combinations on blood lipid and atherosclerosis index (AI) in rats with hyperlipemia, so as to make a preliminary screening for the optimal acupoints combination for hyperlipemia.</p><p><b>METHODS</b>One hundred and five clean-grade SD male rats were randomly divided into 9 groups, including a normal group, a model group, a Quchi group, a Zhongwan group, a Fenglong group, a Quchi+Zhongwan group, a Quchi+Fenglong group, a Zhongwan+Fenglong group and a Quchi+Zhongwan+Fenglong group (three acupoints group), 17 rats in the normal group and 11 rats in the rest groups. The normal group was fed with normal diet, while the rest groups were fed with high-fat diet for 3 weeks to prepare the hyperlipemia model. All the rats were given unlimited water. After the establishment of model, the normal group was fed freely without any treatment; the model group was bundled and immobilized everyday; the rest groups were bundled, immobilized and treated with electroacupuncture at corresponding acupoints with disperse-dense wave, 20 min per time, once a day. After 4 weeks, the blood examples were collected from abdominal aorta to measure the total cholesterol (TC), triglyceride (TG), high-density lipoprotein (HDL-C) and low-density lipoprotein (LDL-C), and analyzed the AI in each group.</p><p><b>RESULTS</b>After the treatment, TC, TG, HDL-C, LDL-C and AI in each acupuncture group were all lower than those in the model group (P<0.05, P<0.01). Compared with single acupoint group and the Quchi+Zhongwan group, the content of TC in the three acupoints group was lower (P<0.01). The differences of content of TG among each acupuncture group were not significant (all P>0.05). Compared with the rest 6 acupuncture groups, the content of HDL-C and AI in the three acupoints group were significantly different (all P<0.05). The content of LDL-C in the three acupoints group was decreased as compared with the Quchi group and the Zhong-wan group.</p><p><b>CONCLUSION</b>The electroacupuncture at "Quchi" (LI 11), "Zhongwan" (CV 12) and "Fenglong" (ST 40) has more advantages on regulating the content of HDL-C and LDL-C as well as improving AI in hyperlipemia rats, and it has superior effects on blood lipid metabolism.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the correlation between the morbidity of pneumonia and meteorological factors in children from Huhhot, in order to provide a basis to prevent and decrease the morbidity of childhood pneumonia.</p><p><b>METHODS</b>A total of 5087 hospitalized children with pneumonia from Huhhot between January 2004 and December 2009 were enrolled. The Circular Distribution method was applied to analyze the seasonal characteristics of the morbidity of pneumonia. The Linear Stepwise Regression Analysis was applied to investigate the relationship between the morbidity of childhood pneumonia and meteorological factors.</p><p><b>RESULTS</b>The morbidity of childhood pneumonia displayed an obvious seasonal trend. Childhood pneumonia was common in winter and spring and its peak morbidity was noted in March. The higher morbidity of pneumonia was related to low air temperature, high air pressure, low precipitation, low humidity and high wind velocity.</p><p><b>CONCLUSIONS</b>Meteorological factors affect the morbidity of childhood pneumonia in Hohhot, and should be considered in the prevention of the disease.</p>

ABSTRACT

Objective To study the effect of lipoic acid on apoptosis of neurons during experimental neuronal aging. Methods The mouse neuroblastoma cells with serum-free culture were served as experimental neuronal aging models.The cells were randomly divided into control group(serum-free culture)and experimental group(100?mol/L lipoic acid in serum-free culture).After cultivating for 5,10 and 15 d,the cell survival rates were detected by MTT method,and the cell apoptosis rates were quantitatively determined using FACS with Annexin V/PI double staining.Results Compared with those in control group,the cell survival rates in experimental group significantly increased[(212.48?12.26)%vs(100.00?14.80)%,(200.55?28.33)%vs(75.45?10.79)%and(96.81?14.50)%vs(28.54?3.14)%,respectively for 5, 10 and 15 d after cultivation](P

ABSTRACT

Objective To investigate the characteristics of cognitive function impairment in patients with epilepsy. Methods Forty-five epilepsy patients and twenty age-,gender-,and education-matched healthy controls were submitted to a wide neuropsychological battery,including Raven's standard progressive matrices,auditory verbal memory test,logical memory test,visual memory test,Stroop color words test,trail making test,verbal fluency test,Boston naming test,math and complex figure test,etc. Results The epilepsy patients performed significantly worse than the controls in the verbal delayed memory,verbal short memory,cued recall,immediated recall of logical memory test,visual spatial memory,attention,information processing speed test,word finding difficulty and logical reasoning.Conclusion There exists impairment in various cognitive domains in patients with epilepsy,and the main impaired areas are memory,executive functions and logical reasoning.

ABSTRACT

Objective To study the effects of a rotating magnetic field(RMF)on Li pilocarpine-in- duced seizure activity and the expression of mGluR1 and mGluR5 in the hippocampus.Methods Thirty rats were divided into 5 groups.Each rat in the model(M),short treatment(ST)and long treatment(LT)groups was treated with intra-peritoneal injections of lithium chloride(60 mg/kg),followed by an intra-peritoneal injec- tion of pilocarpine(35 mg/kg)24 h later.The rats in the ST group were exposed to 20 mT RMF for 20 min ev- ery day for 3 d before seizure induction,while the rats in LT group were exposed to the same RMF for 8 d.The latency,severity and duration of seizure,as well as accompanying symptoms and electroencephalogram data, were recorded,and the expression of mGluR1 and mGluR5 was calculated using an electrophoretic imaging anal- ysis system.Results The duration,times and accompanying symptoms of seizure were significantly decreased in the LT group.The mGluR1 mRNA level and mGluR1/mGluR5 ratio in the M group were markedly increased, but the mGluR5 mRNA level was obviously decreased,while the expression of mGluR1 in the ST and LT groups was decreased,and mGluR5 was increased.Conclusions Seizure activity in rats can be inhibited by 20 mT RMF,and the expression of mGluRl and mGluR5 in the hippocampus of rats suffering seizures can be markedly influenced by longer-term RMF.

ABSTRACT

Objective To analyze the etiology and clinical feature of pi t uitary apoplexy.Methods The clinical data from 18 cases of pitui tary apoplexy were analyzed retrospectively.Results Main symptom s were headache in 12 cases and decreased visual acuity in 12 cases.Positive si ghs were temporal hemianopisia in 6 cases,unequal pupil in 3 cases,oculomotor ne rve(Ⅲ) paralysis in 2 cases and abducent nerve (Ⅵ) paralysis in 2 cases.Imagin g examinations showed CT positive finding in 4 cases and MRI positive finding in 6 cases.Tumor diameter was longer than 2 cm in 13 cases and less than 2 cm in 5 cases.Chromophobe pituitary tumor was found in 11 cases.All cases received surg ical operation.Conclusion Clinical features of pituitary tumor a poplexy are atypical,of which headache and visual acuity decrease are the main s ymptoms.Most cases occur suddenly or primary symptoms are deteriorated suddenly. CT and MRI positive findings are helpful for diagnosis.

ABSTRACT

PKS gene was screened by PCR from thirty strains of spone-associated bacteria including twenty-one actinomycetes isolated from Craniella anstrialiensis and nine bacillus isolated from Dysidea avara in the South China Sea.As a result,a 669 bp KS domain gene was successfully amplified from Bacillus C89.BLAST analysis showed that the KS domains were most closely related to the KS sequences of Bacillus subtilis subsp.subtilis str.168 with 96% similarity.Phylogenetic analysis demonstrated the KS domain belong to trans-AT KS domains.This study demonstrated the existence of PKS gene in bacteria associated with sponge Dysidea avara for the first time,and provided proof for the hypothesis that sponge-associated bacteria are perhaps the true producers of many novel bioactive compounds in sponge.Meanwhile,this study lays a basis for the microbial screening for polyketide compounds production.
